The pharmaceutical industry relies on HPLC through all stages of drug development, from drug discovery to manufacture. Most commonly, bench-scale HPLC instruments are used, usually located in dedicated analytical laboratories, meaning that samples need to be transferred from the work site. There is therefore considerable potential for the development and application of a miniaturised, portable HPLC system that can be deployed at-line or on-line in a range of situations within the pharmaceutical industry. This project, which involves collaboration between a university research centre (University of Tasmania), two major pharmaceutical companies (Pfizer and GlaxoSmithKline), and a scientific instrument developer (Trajan Scientific and Medical) is exploring these possibilities. A miniaturised, briefcase-sized gradient elution capillary HPLC platform has been developed using syringe pumps and single wavelength UV detection using a light-emitting diode (LED) light source with an end-capillary extended optical pathlength (1.2 mm) flow-cell in a 3D-printed housing. This basic platform will be described in detail, together with a range of additions designed to improve ease of application and to increase detection selectivity and sensitivity. These additions include on-line sample dilution, direct interfacing of swab sampling, multi-wavelength light sources (including a miniaturised deuterium lamp and a multi-LED fibre optic light source), spectral scanning capabilities via a miniature photodiode array detector, and direct interfacing with a portable mass spectrometer. In the current configuration, the LC system itself is 30 x 45 x 10 cm and weighs 7 kg. Projects are under way to adapt these HPLC and LC-MS platforms to specific applications in the pharmaceutical industry, including on-line reaction monitoring in drug development and quality control of active ingredients and impurities in continuous drug synthesis systems.
